Mesenchymal stem cells (MSCs) possess potent immunoregulatory properties and are a source of numerous growth and trophic factors. These properties make them promising candidate for ocular surface regeneration. It was demonstrated that MSCs can express markers of corneal epithelial cells and keratocytes under selected conditions. In previous study, we found that insulin-like growth factor-I (IGF-I) supports the differentiation of limbal stem cells into corneal-like cells. In the present study, we tested differentiation of mouse bone marrow-derived MSCs into corneal epithelial cells using the extract from the cornea and IGF-I. The results show that MSCs cultured in the presence of corneal extract and IGF-I efficiently differentiate into corneal-like cells. The differentiated cells possess characteristics of corneal epithelial cells and keratocytes and at the same time maintain MSC properties.
