The additive effect of endothelin-1 blockade to renin-angiotensin blockade was evaluated in rats with ablation nephrectomy. While RAS blockade normalized blood pressure (BP), improved survival, decreased cardiac hypertrophy and proteinuria as well as tissue angiotensin II and ET-1 levels, ETA receptor blockade only partially improved survival rate, reduced BP, attenuated the development of cardiac hypertrophy and transiently reduced proteinuria. No additive cardio- and renoprotective effects of ETA and RAS blockade were noted at the end of the study suggesting that the RAS system is the main target for antihypertensive treatment in models with inappropriately activated RAS
