In recent years, cell replacement strategies, together with endogenous stem cell stimulation within the adult central nervous system, has been considered as a promising therapeutic approach to compensate for destroyed neural pools following various neurological disorders. Therefore, a great deal of effort has been going towards establishing the factors which control cellular survival, proliferation and differentiation. Nitric oxide (NO), a diffusible intracellular, intercellular messenger and powerful mediator in the inflammatory cascade in the mammalian nervous system, has been shown to affect embryonic as well as adult neurogenesis. It has been confirmed by several groups that at physiological concentrations NO derived from the neuronal isoform of NO synthase (nNOS) in nitrergic neurons is a negative regulator of stem/progenitor cell pool proliferation and is most likely needed for initiation of differentiation.
