    Purinergic P2X receptors are ubiquitous, ligand-gated cationic channels activated by extracellular ATP. The P2X3 receptors, which are involved in nociception, are known for their very fast and complete desensitization, which persists for many minutes. Structural modeling followed by experimental validation of predictions suggested conserved tyrosine 37side chain is needed for the stabilization of the closed receptor state while kinetic modeling specifies that lack of Y37 results in destabilization of closed desensitized states
