Psychiatric diseases are associated with elevated levels of pro-inflammatory cytokines in affected patients. Cytokines IL-1 and TNF- enhance the production of serotonin transporter (5-HT) thus, antidepressants could mediate their effect by downregulation of these cytokines. Therefore, we studied the influence of ketamine in male Wistar/Hann rats on IL-1 and TNF- and 5-HT activity in the blood and brain. Both cytokines increased up to 140% and 150% only in control animals, not in the ketamine group. Ketamine administration led to lower 5-HT levels in the hippocampus compared to controls (t=2.218, p<0.05). The hippocampal 5-HT levels correlated negatively with the basal TNF- and positively with the absolute and relative change of this cytokine in the whole sample, which suggests inhibition of IL-1 and TNF-activity. The mechanisms responsible for the interaction between the pro-inflammatory pathway, 5-HT and antidepressant effect of ketamine remain unknown
