Optical fibers have been used in the past to image in locations deep in the scattering tissue which are not accessible through optical microscopy. Recently, the endeavor to minimize tissue damage has motivated the use of multi-mode fibers which allowed for high-resolution imaging with significantly smaller cross-section than single-mode fibers or GRIN lenses. Only recently the first successful functional imaging of neuronal calcium in vivo has been demonstrated in the mouse brain. These efforts represent a huge leap forward in MMF-based endoscopy, yet the quality of the detected functional calcium signals in vivo is still far from the state-of-the-art standards set by confocal and multi-photon microscopy. We introduce here computational and technological advancements to improve the quality of fiber-based imaging potentially important for brain endoscopy.
