    In our study we applied both traditional and advanced microscopic imaging techniques to study testate amoebae (TA) shell and cell morphology. When compared with scanning electron microscopy (SEM) or environmental SEM (ESEM), confocal laser scanning microscopy (CLSM) and two-photon excitation microscopy (TPEM) offered several additional advantages for investigations requiring information of morphological characteristics, useful for taxonomical and ecophysiological research: (1) CLSM and TPEM allow imaging of inner structures of TA in contrast to SEM or ESEM. (2) CLSM and TPEM can simplify and refine studies of morphology. (3) CLSM and TPEM allow us to obtain data that can be used for stereological estimations of volumes of TA. (4) Fluorescence markers allow us to see structures not visible in transmitting light
