    The ubiquitin-proteasome system regulates many cellular processes via proteasomal degradation of ubiquitinated proteins. Ubiquitin C-terminal hydrolase-L1 (UCHL1), an abundant protein in mammalian oocytes, has hydrolytic activity as a monomer. In its dimeric and oligomeric form, UCHL1 acts as an ubiquitin ligase. Recently was found that, deficiency in UCHL1 coincides with polyspermic fertilization; however, the role of UCHL1 in such process remains unclear. After in vitro fertilization, a high rate of polyspermy in bovine zygotes was induced by UCHL1 inhibitors and decreased level of the monomeric as well as polymeric ubiquitin pool was detected. In addition, presumptive UCHL1 oligomers were formed after UCHL1 inhibition with subsequent increase of K63-linked polyubiquitin chains in oocytes. Moreover, both migration of cortical granules (CGs) toward the cortex during oocyte maturation and fertilization-induced extrusion of CGs were impaired after UCHL1 inhibition.
