Proteins were digested from barley leaves, then separated using ID gel electrophoresis and in-gel digested with trypsin. Peptide masses were adjusted through MALDI-TOF and then put into the database searching program. This method is not always successful because the success depends mainly on presence of the protein in the database. In the case of barley only about 20
